Definition

Agar base is enriched with blood for the in-house preparation of blood agar. It is an enriched medium extensively used for cultivation and isolation of microorganisms.

Soybean Casein digest Agar, TSA, CASO Agar, Tryptone Soya Agar.

Instructions for use

  • ​Follow Standard Operational Procedures for Blood Agar plates preparation.
  • Ensure aseptic conditions during medium preparation.
  • Once prepared store Blood Agar plates refrigerated 2-8ºC. Use no longer than 4 weeks after preparation.
  • ​Before use, perform internal quality control. To check for sterility, incubate few plates from each new batch for 2 days at 36ºC.
  • Add defibrinated blood when agar has cooled down to avoid haemolysis.

Storage

  • At room temperature below 30°C
  • Store in dry conditions in well sealed receptacles far from oxidizing substances
  • Shelf life: 6 months

MSF requirements

Agar Base is used for in-house preparation of Blood Agar plates. Those are used to check if broth from positive MGIT cultures contains contaminants.

It is recommended to purchase blood agar plates already prepared if in-house preparation of blood agar plates is not a routine laboratory procedure. Refer to your laboratory advisor.
