Definition

Columbia agar is highly nutritious medium for the isolation of non fastidious and fastidious microorganisms when supplemented with sheep blood. The medium is used as an efficient base for preparation of other media as Chocolate or Blood Agar. It is a culture medium used for the growth and the isolation of germs, commonly found in various types of samples.

Components

Approximate formula after reconstitution of the medium (1 liter):

  • Pancreatic digest of casein 10 g
  • Meat peptic digest 5 g
  • Yeast extract 5 g
  • Heart pancreatic digest 3 g
  • Corn starch 1 g
  • Sodium chloride 5 g
  • Agar 13.5 g

Technical specifications

  • Dehydrated medium, powder.
  • Dehydrated appearance: Beige, free flowing, homogeneous
  • Prepared appearance: Plain-light to medium amber, slightly opalescent to opalescent with fine precipitate. With sheep blood - cherry red, opaque, no hemolysis

Instructions for use

  • Follow the recommendations of the package insert.
  • Do not use non homogenous media.
  • Properly close the bottle after use.

Precautions for Use

All biological samples must be considered as potentially infectious and handled with the usual precautions (compulsory wearing of gloves, hand washing, etc.).

Storage

  • Follow the recommendations of the manufacturer for the storage.
  • Preserved from humidity.
  • Do not use after expiry date.
  • After preparation, store the media maximum 3 weeks between 2°C and 8°C.

Waste management

After use, sterilize the cultures and all contaminated material. Please contact your watsan referent for advice on proper disposal.
